What are the criteria for COSMIC hotspot mutations?

Hello DepMap team,

I read in Ghandi et al (2019) that cancer hotspot mutations that are labeled as TRUE for isTCGAhotpot are a “subset of variants that were highly recurrently observed in TCGA (in 6 or more TCGA samples).” However, I could not find any documentation regarding what is considered a hotspot for isCOSMIChotspot, can you elaborate on how you define this?

Cheers,
Yuka

Hi Yuka, the isCOSMIChotspot column is based on the column COSMIC_overlapping_mutations from Oncotator annotations. We consider a locus COSMIC hotspot if at least 10 hits are reported for it on the COSMIC database
