My gene of interest doesn’t show to be essential in any of the cell lines tested in Depmap in the CRISPR screens. How do I know that the gene has been efficiently knocked out, and the % of KO? Has it been validated?
Hello! We do not have a measurement of knockout in Achilles or further experimental validation. For your gene of interest, you could look at the reagent level (to see if any of the individual guides targeting your gene show depletion in the logfold change data) or check if the gene scores as a dependency in any of the RNAi or Project SCORE screens (as it is unlikely that the KO of a gene would be poor in three independent libraries).
