Estimation of uncertainty around the dose-response curve

Hi everyone,
I was wondering, how do you estimate the uncertainty around the AUC value for the drug screens?
Some of the cell lines were tested in 2-3 replicates and some in 6, that would influence how certain we are on the curve fit from cell line to cell line. Also those AUCs become less directly comparable in that case.

Would be grateful for any tip!

Best regards,

Katarzyna

Hi Katarzyna,

Today we don’t report the uncertainties around the AUC values and that is something to work on our list for now. But, the problem is beyond the number of replicates since the inherent noise level for each cell line also varies (typically faster cell lines are less noisy). So, unfortunately, we don’t have a clear-cut solution that we can recommend.

If you find any good practice around this issue, your feedback would be very welcome.

Warmly,
Mustafa
